摘要
目的建立不同产地的马齿苋中槲皮素、山奈酚、木犀草素、芹菜素含量测定方法。方法高效液相色谱法,色谱柱ZORBAXSB-C18(150mm×4.6mm,5μm);检测波长:360nm;甲醇-0.2%磷酸(45∶55)为流动相;柱温:30℃;流速:1.0ml/min。结果槲皮素、山奈酚、木犀草素、芹菜素回归方程分别为Y=1504.412X+9.9756,Y=1991.745X+8.6051,Y=567.591X+2.5397,Y=1811.803X+0.3074,在0.0536~0.1877、0.0461~0.1613、1.255~4.393、0.0602~0.211μg/ml范围内线性良好,r=0.9999~0.9999,加样回收率分别为95.7%、96.9%、107%、91.1%,RSD分别为2.99%、3.53%、1.89%、2.81%。3个样品含槲皮素、山奈酚、木犀草素、芹菜素的总和分别为0.176、0.218、0.228mg/g。结论本法为马齿苋提供了分析4种黄酮化合物方法 ,该法简便可行,重复性好。
Objective To establish a determination method for quercetin,kaempferol,luteolin,apigenin in Portulaca oleracea L.from different places.Methods Reverse-phase high performance liquid chromatography menthod was used:the column was ZORBAX SB-C18;detection wavelength was set at 360 nm;menthol-0.2%phosphonic acid(45∶55)was used as the mobile phase;column temperature was 30℃ and the flow rate was 1.0 ml/min.Results The regression equation of quercetin,kaempferol,luteolin,apigenin was Y=1504.412X+9.9756,Y=1991.745X+8.6051,Y=567.591X+2.5397,Y=1811.803X+0.3074 respectively ;the linear range was(5.36×10-2-18.77×10-2)μg/ml,(4.61×10-2-16.13×10-2)μg/ml,(125.5×10-2-439.26×10-2)μg/ml,(6.03×10-2-21.10×10-2)μg/ml respectively ;correlation coefficents were r=0.9999-0.9999 and the average recovery was 95.7%,96.9%,107%,91.1%;RSD 2.99%,3.53%,1.89%,2.81% respectively.The sum contentions of quercetin、kaempferol、luteolin、apigenin in three samples were 0.176,0.218,0.228 mg/g respectively.Conclusion This menthod is simple,effective and suitable for analysis of the above flavonids in portulaca oleracea L.
