一测多评法测定连翘中多种不同类型成分的含量 被引量:47

摘要 目的采用一测多评法同步测定连翘中4个主要成分的含量,重点探讨不同类型化合物之间开展一测多评研究的准确性和可行性。方法采用HPLC-DAD,计算指标性成分芦丁与连翘酯苷A、连翘苷、连翘酯素在275 nm处的紫外相对校正因子(relative correction factor,RCF),并将该校正因子应用于连翘药材中上述4种成分的含量计算,实现一测多评;同时采用外标法测定药材中芦丁、连翘酯苷A、连翘苷、连翘酯素的含量,比较计算值与实测值的差异。结果 11批连翘药材中3个不同类型4种成分的含量可以用一测多评法进行测定,其计算值与实测值间无显著差异。结论在缺少对照品的情况下,以外标法测定芦丁,利用相对校正因子实现对连翘酯苷A、连翘苷、连翘酯素的含量测定是快速的、可行的,一测多评法可以用于连翘中不同类型成分间的定量评价研究。 OBJECTIVE To investigate the quantitative analysis of multi-components by single marker(QAMS) for the different types of compounds in the fruit of Forsythia suspensa.METHODS A HPLC method was developed as QAMS to determine the four main components(rutin,forsythoside A,forsythin and forsythigenin) in F.suspensa.Rutin was selected as the internal reference substance,and 3 relative correction factors(RCF) to rutin were calculated.The contents of the four components in 11 batches of samples,collected from different areas,were determined by both external standard method and QAMS.The method was evaluated by comparison of the quantitative results between the two methods.RESULTS No significant differences were found between the quantitative results of four components in 11 batches of F.suspensa determined by the two methods.CONCLUSION QAMS is feasible for determination of certain different types of compounds,and the developed method can be used for quality control of F.suspense.
