摘要
目的探讨重组腺病毒载体介导的微小RNA-21(miRNA-21)反义寡核苷酸(ASOND)对人膀胱癌细胞株T24生长的影响。方法构建反向插入miRNA-21基因的重组腺病毒载体(rAV-miRNA-21),并用反义rAV-miRNA-21转染T24,即病毒载体用药组。采用噻唑蓝(MTT)法和原位末端标记(TUNEL)法检测T24细胞的增殖和凋亡情况。同时设对照组和空病毒组作为对比。结果 rAV-miRNA-21用药组、对照组和空病毒组T24细胞的增殖率分别为(37.1±6.8)%(、95.7±5.8)%(、86.3±7.5)%;凋亡率分别是(31.6±4.7)%(、8.4±2.9)%(、7.3±4.6)%。用药组的增殖率和凋亡率分别与对照组和空病毒组相比较,差异均有统计学意义(P<0.01),而对照组与空病毒组之间差异无统计学意义(P>0.05)。结论反义miRNA-21重组腺病毒载体转染T24细胞可以抑制肿瘤细胞增殖,增加细胞凋亡,进而抑制膀胱癌细胞生长。
Objective To explore miRNA-21 antisense oligonucleotide mediated by recombinant adenovirus vector affect human bladder cancer cell line T24 growth.Methods To construct inverted insertion miRNA-21 gene recombinant adenovirus vector(rAV-miRNA-21),and use rAV-miRNA-21 transfect T24,that is viral vector medication group.Adopting MTT and TUNEL detect proliferation and apoptosis status.Simultaneously set up control group and take vacancy viral group as comparison.Results The T24 growth rates of rAV-miRNA-21 group、control group and vacancy viral group are(37.1±6.8)%,(95.7±5.8)% and(86.3±7.5)%,the apoptosis rates are(31.6±4.7)%,(8.4±2.9)% and(7.3±4.6)%.Compare proliferation and apoptosis of rAV-miRNA-21 group miRNA-21 group with control group and vacancy viral group,difference has statistical significance(P0.05),but between control group and vacancy viral group difference has nonsignificance(P0.05).Conclusion Antisense miRNA-21 recombinant adenovirus vector transfect T24 could inhibit proliferation of tumor cell and increase tumor cell apoptosis,and so inhibit bladder cancer cell growth.
