摘要
目的探讨血管紧张素受体拮抗剂氯沙坦(ARB)对血管紧张素受体亚型1(AT1)、血管紧张素受体亚型2(AT2)、血管紧张素转化酶(ACE)、血管紧张素原(AGT)基因表达的影响。方法本试验应用阿霉素(ADR)复制肾衰大鼠模型,采用RT-PCR检测AT1、AT2、ACE、AGT基因表达。结果肾衰组肾脏AT1表达上调,AT2降低,氯沙坦组转为AT1下调,AT2表达接近对照组水平;与对照组相比,肾衰模型AGT水平增高,而给予氯沙坦后其水平降低;ACE在肾衰时表达上调,给氯沙坦后降低但仍高于对照组水平。结论氯沙坦可从调节血管紧张素II(Ang II)生成及受体水平角度对抗AngⅡ的作用,从而避免肾损伤。
Objective To explore the mRNA expression of AT1,AT2,ACE and AGT was determined with RT-PCR method.Methods In this study,after the rat model of renal failure was established by administrating with ADR.Results RT-PCR results showed that adriamycin inducted that upregulation of AGT,ACE and AT1,and down-regulation of AT2,but the down regulation of AGT,AT1 when adriamycin administration.Conclusion ARB can competitively inhibit the acitivity of ANG II through binding with AT1,or down regulate the expression of ANG II receptor,it can protect the kidney from the injury.
