摘要
目的观察三七总皂甙(PNS)对实验性糖尿病性心肌病大鼠心肌超微结构及心肌酶相关指标的影响。方法将60只SD大鼠随机分为正常组,模型组,PNS低、中、高剂量组,达美康组,每组10只。造模成功后,正常组、模型组按容积10mL/kg予蒸馏水灌胃;PNS低、中、高剂量组按生药50、100、150mg/kg,容积10mL/kg予PNS溶液灌胃;达美康组按生药20mg/kg,容积10mL/kg予达美康缓释片溶液灌胃。各组均每日1次灌胃,连续4周。4周后检测各组大鼠空腹血糖(FPG),光镜、电镜观察各组大鼠心肌超微结构,免疫组织化学法检测各组大鼠心肌酶相关指标碱性磷酸酶(ALP)、三磷酸腺苷酶(ATPase)含量及琥珀酸脱氢酶(SDH)活性。结果 PNS低、中、高剂量组及达美康组大鼠FPG与模型组比较差异均有统计学意义(P<0.01),PNS低、中、高剂量组及达美康组低于模型组;PNS低、中、高剂量组光镜、电镜下可见心肌细胞超微结构及心肌酶相关指标明显改善;PNS低、中、高剂量组大鼠ALP、ATPase含量及SDH活性与模型组、达美康组比较差异均有统计学意义(P<0.01,P<0.05),PNS低、中、高剂量组高于模型组及达美康组。结论 PNS治疗糖尿病性心肌病大鼠疗效确切,对心肌具有保护作用。
Objective To observe the ultrastructural changes of myocardium in diabetic cardiomyopathy rat affected by panax notoginseng saponins (PNS).Methods 60 SD rats were randomly divided into control group, model group, Low-, middle-, high-dose PNS groups and Diamicron group, 10 rats in each group. After model establishment, distilled water, 10 mL/kg Low-, middle-, high-dose PNS (50, 100,150 mg/kg) and 10 mL/kg Diamicron retard tablets (20 mg/kg) were lavaged in different groups. The course was four weeks in each group. After a course FBG in rats was determined. And cardiac ultrastructure changes were observed by light microscope and electron microscope. ALP, ATPase and SDH were measured by immunohistochemical method. Results There was significant difference between Low-, middle-, high-dose PNS groups and Diamicron group and model group on FPG (P<0.01). The cardiac ultrastructure was obviously improved under light microscope and electron microscope in Low-, middle-, high-dose PNS groups. The activities of ALP, ATPasa and SDH in Low-, middle-, high-dose PNS groups were higher than those in model group and Diamicron group (P<0.01,P<0.05). Conclusion Panax notoginseng saponins has definite effect on diabetic cardiomyopathy rat, and can protect myocardium.
