摘要
对毛蚶、魁蚶和泥蚶核糖体DNA转录间隔区(ITS区)进行限制性片段长度多态性(RFLP)分析发现,6种限制性内切酶(HaeⅢ,HhaⅠ,HpaⅡ,RsaⅠ,Sse9Ⅰ和TaqⅠ)均能将泥蚶与毛蚶及魁蚶区别开来,但未发现毛蚶和魁蚶间特异性的RFLP标记。对酶切图谱进行的数理分析结果显示,毛蚶与魁蚶之间的净遗传距离Pnet值为0,而泥蚶与毛蚶、泥蚶与魁蚶之间的Pnet值均为0.0797;泥蚶群体内检测到5种酶切复合单倍型,核苷酸多样性指数π值为0.0008外,而毛蚶和魁蚶群体内为相同的1种复合单倍型,π值均为0。
In this paper,RFLP analysis was conducted in ribosomal DNA internal transcribed spacers (ITS) of Arca subcrenata,Arca inflata and Arca (anadara) granosa. The results showed that A. granosaa and A. subcrenata could be differentiated using restrictive enzymes of HaeⅢ,HhaⅠ,HpaⅡ,RsaⅠ,Sse9Ⅰand TaqⅠ,as well as A. granosaa and A. inflata. No specific RFLP marker was observed between A. subcrenata and A. inflata. Pnet value between A. subcrenata and A. inflata was zero,while Pnet values between A. granosa and the other two species were both 0. 079 7. It was observed that there were 5 composite haplotypes in A. granosa and the nucleotide diversity index (π value) was 0. 000 8,while one haplotype was observed in A. subcrenata and A. inflata and π values were both zero in these two species.
