摘要
肝素(Heparin,Hep)为葡糖胺聚糖,是蛋白多糖的一种,人体内由肥大细胞分泌而自然存在于血液中。它具有广泛的生物学功能,临床上是预防血栓形成和治疗急性静脉血栓的重要药物。肝素的药效与剂量密切相关,因此研究低剂量及超低剂量肝素的定量分析方法有重要意义。
A new resonance Rayleigh scattering (RRS) method for the determination of heparin with Acriflavine hydrochloride (AH) has been developed. In pH 5.0 BR buffer solution, heparin can react with Aeriflavine hydrochloride to form an ionassociation complex,which will result in great enhancement of RRS and the appearance of a new RRS spectrum. The maximum RRS peak is located at 505nm. The intensities of RRS are proportional to the concentration of heparin in the range of 0. 080 - 1.20mg/ L. The method has very high sensitivity and the detect limit for the heparin is 0. 022mg,/L. The method can be applied to the determination of trace amounts of heparin with satisfactory results. The recoveries are 97.0 - 104.5%.
