尿毒清颗粒改善链佐星-弗氏完全佐剂诱导的糖尿病肾病模型肾脏功能的研究 被引量:6

摘要 目的:观察尿毒清颗粒(大黄、黄芪、甘草、茯苓等)对链佐星-弗氏完全佐剂诱导的实验性糖尿病肾病(diabetic nephropathy,DN)大鼠肾脏功能的改善作用及可能的机制。方法:以链佐星-弗氏完全佐剂辅以高脂饮食制作DN大鼠模型,检测尿液指标、血清生化指标,并进行病理、免疫组织化学分析。结果:尿毒清颗粒可明显减少DN大鼠的进食量,减少24h尿量,降低24h总蛋白和白蛋白排出量和红细胞和肾脏匀浆中山梨醇含量;降低血清中血糖、MDA、肌酐、尿素氮、糖基化终产物(AGEs)水平,提高SOD水平;提高肌酐清除率;明显降低系膜区增厚、基质增多等病理变化;减少肾小球IV型胶原的表达。结论:尿毒清颗粒明显改善DN模型的肾脏功能。 AIM : To investigate the effect of Niaoduqing Granule ( Radix et Rhizoma rhei, Radix astragali, Ra- dix et Rhizoma glycyrrhizae, etc) on renal function of rats with diabetic nephropathy (DN) and the mechanisms in- volved. METHODS: A modified DN rat model (multiple injections of low-dose streptozotocin and complete Fre- und~ adjuvant) was used and the related indexes were detected. RESULTS: Food intake, 24-hour urine volume, proteinuria, albuminuria and sorbitol concentration in red blood cells and kidney homogenate were attenuated by treatment with Niaoduqing Granule. Levels of blood glucose, malonaldehyde, creatinine, blood urine nitrogen, advanced glycation end-products in the serum were decreased, whereas superoxide dismutase activity and creatinine clearance rate were increased in the Niaoduqing-treatment group. Furthermore, Niaoduqing Granule significantly decreased glomerular extracellular matrix expansion & accumulation and collagen type IV expression in the kidneys of DN rats. CONCLUSION: Niaoduqing Granule could improve the renal function of DN rats.
