摘要
目的:研究那如-3丸对大鼠佐剂性关节炎大鼠的治疗作用及部分机制。方法:用弗氏完全佐剂诱导大鼠AA模型;足容积法测量继发性足肿胀度;比色法测定脾淋巴细胞增值反应;用ELISA法测定IL-1、TNF-a、PGE2水平;用比色法测定NO、SOD、MDA含量。结果:致炎后第15天,大鼠继发性关节炎出现,致炎后第8天开始预防给予不同剂量的那如-3丸,连续15d,从致炎后第15天起开始各给药组足跖容积和足跖肿胀度明显下降,同时大鼠关节组织的IL-1、TNFa、PGE2、NO的含量明显下降,但那如-3对SOD、MDA含量无明显影响。结论:那如-3丸对AA大鼠继发性炎症具有治疗作用,起作用机制可能与调节机体异常的免疫功能和维持细胞因子网络平衡有关。
AIM： To study the therapeutic effect of Naru-3 Pill on adjuvant arthritis （ AA ） in rats and its meehamisms. METHODS： Freund＇s complete adjuvant （FCA） was used to induce AA in rats. Secondary paw swelling of AA rats was measured With volume meter. Splenic lymphocyte proliferation response was estimated by colorimetry; IL-1, TNF-α and PGEa-production by enzyme linked immunosorbent assay （ELISA）; levels of NO, SOD,MDA-produetion by eolorimetry. RESULTS： The secondary inflammation of AA rats appeared on the 15th day after injection of FCA. 8 d after inflammation, different doses of Naru-3 Pill were offered to prevent inflamma- tion for 15 d. From the 15th day, paw volume and paw swelling of rats in drug-treated groups significantly decreased,while the level of IL-1, TNFa, PGE2, NO in rat joint tissue was significantly decreased, but Naru-3 Pill showed no significant effect on the level SOD, MDA. CONCLUSION： The results suggested that Naru-3 Pill had therapeutical effect on AA rats. Its mechanisms may be related to adjusting abnormal immune function in AA rats and keeping the balance of cytokine network.
