摘要 针对当前对生物酶降解纤维素作用机理研究不足,文章通过DNS比色法,考察不同纤维素酶对纤维素的降解过程,并考察了温度、pH值和表面活性剂等外界条件对生物酶降解性能的影响。研究发现,温度对纤维素酶的活性的影响规律是先升高后降低的趋势,50℃是纤维素酶活性最高的温度;pH升高对纤维素酶活性起着抑制作用,pH=4的偏酸性环境最适合纤维素酶解。表面活性剂十二烷基硫酸钠、十二烷基苯磺酸钠和TritonX-100对纤维素酶的水解活性均有抑制作用。文章的结论将有助于指导纤维素酶的现场应用。 This paper has studied the enzyme degradation of cellulose derivatives by different cellulose enzymes with DNS color comparison,and then,the effects of ambient conditions such as temperature,pH value and surfactants on the enzyme degradation performance also were studied.The results showed that,temperature is an important factor for the enzyme degradation,its rule is rising to maximum and then declining,and the activity of enzyme is maximum at 50℃.The pH value affects the enzyme degradation greatly,whose activity is maximum at pH = 4 partial acid environment.The surfactants including Sodium Dodecyl Sulfate,Sodium Dodecyl Benzene Sulfonate and Triton X-100 have inhibiting action for the enzyme activity.In summary,the results may be helpful in guiding the application of cellulose enzymes.
