摘要
以自制牛骨粉为原料,水解度为指标,选用Ⅱ型胶原酶与枯草中性蛋白酶对牛骨粉进行分步水解,采用L9(34)正交试验设计优化2种酶水解牛骨粉的工艺条件。结果表明,Ⅱ型胶原酶与枯草中性蛋白酶先后对牛骨粉酶解的水解效果比单一的Ⅱ型胶原酶效果好。2种酶水解的适宜工艺条件:先用Ⅱ型胶原酶在37℃条件下恒温震荡水解3 h,再用枯草中性蛋白酶在50℃条件下恒温震荡水解3 h,固液比1∶4,酶用量0.3%,2种酶用量比1∶1,pH 7.0,其水解度达16.65%,胶原多肽的分子量主要分布在30~50 kD。
The technological condition of collagen polypeptide from hydrolyzed bovine bone powder by collagenaseⅡ and bacillus subtilis neutral protease was optimized by L9(34)orthogonal test.The results showed that the hydrolyzing degree of collagenaseⅡand bacillus subtilis neutral protease was better than that of collagenaseⅡ,the hydrolyzing degree of the bovine bone powder hydrolyzed by collagenaseⅡ at constant 37℃ for 3 h,and then hydrolyzed by bacillus subtilis neutral protease again at constant 50℃ for 3 h under the condition of 1∶4 solid/liquid ratio,0.3% enzymatic dosage,1∶1 collagenaseⅡ/bacillus subtilis neutral protease and pH 7.0 was 16.65% and the molecular weight of collagen polypeptide was 30~50 kD.
