非酒精性脂肪性肝病兔胰岛素水平变化及其机理探讨 被引量:19

Changes of plasma insulin level in rabbits with nonalcoholic fatty liver disease and its mechanism

摘要 目的观察高脂血症(hyperlipidemia)致非酒精性脂肪性肝病(NAFLD)兔胰岛素(insulin,Ins)水平变化,探讨变化机理。方法 40只日本大耳白兔随机分为标准NAFLD模型组(A组)、半量NAFLD模型组(B组)、空白对照组(C组)。A组给予高脂饲料160g/(兔·d),B组给予高脂饲料80g/(兔·d)+普通饲料80g/(兔·d),C组给予普通饲料160g/(兔·d)。饲养周期13w。实验前后采集血浆标本,常规检测实验前后Ins、甘油三酯(TG)、总胆固醇(TC)、血糖(BS)水平;肝脏组织HE染色,光镜观察肝脏病理学变化。结果 (1)Ins:C组饲养前后无差异;B组饲养后Ins升高(P<0.05);A组饲养后Ins水平升高(P<0.01),且高于B组(P<0.05)。(2)BS:饲养后A组、B组血糖显著升高(P<0.05或P<0.01),且A组高于B组(P<0.05)。(3)TC、TG:饲养后A组、B组与饲养前比较升高(P<0.05或P<0.01),且A组高于B组(P<0.05)。(4)肝脏病理学:A组重度NAFLD,B组轻度~中度NAFLD,C组正常肝脏组织。结论高脂血症致NAFLD兔Ins水平升高,且与NAFLD程度有关。 Objective To observe the changes of insulin level caused by hyperlipidemia in rabbits with nonalcoholic fatty liver disease (NAFLD) , and to explore its mechanism. Methods 40 Japanese white rabbits were randomly divided into standard model of NAFLD group (group A) ,half dose of NAFLD group (group B) and blank control group (group C). Group A was given 160 g of high fat diet daily per rabbit;group B were given 80 g of high fat diet plus 80 g of normal diet daily per rabbit;group C was given 160 g of normal diet daily per rabbit. All animals were fed for 13 w. Their plasma samples were collected before and after treatment for the measurement of the levels of insulin, triglyeride (TG), total cholesterol (TC) and blood sugar (BS). In addition, hepatic tissues were stained with HE and the pathologic changes were observed under light microscope after treatment. Results Insulin level in group A and group B significantly increased after feeding(P 〈0.05 or P 〈0. 01 ) ,and it was higher in group A than that in group B after feeding (P 〈0.05) ,whereas group C had no significant difference between before and after feeding. BS level in group A and group B increased significantly after feeding ( P 〈0. 05 or P 〈 0. 01 ), and group A was higher than group B ( P 〈0. 05 ). TC and TG levels in group A and group B increased significantly after feeding (P 〈 0.05 or P 〈 0.01 ), and group A was also higher than group B( P 〈 0. 05 ). Hepatic pathologic findings indicated that NAFLD was severe in group A and mild to moderate in group B while group C was normal. Conclusion Hyperlipidemia can cause an increase of plasma insulin level that is related to the degree of NAFLD in rabbits.
