摘要
目的:探讨大鼠发育过程中,SSeCKS(Src抑制的蛋白激酶C的底物)在脑组织中的表达变化及意义。方法:通过Western blot,免疫组织化学法检测SSeCKS在脑组织中表达的时空变化及细胞定位。结果:SSeCKS在大鼠脑发育过程中存在明显的表达变化,在胎鼠和生后早期的SD大鼠中表达水平较高,尤以生后1周明显,成年以后显著降低,4周时最低。脑分区检测显示端脑、海马和间脑符合全脑检测结果。DAB显色示SSeCKS在脑组织中呈点状散在分布,免疫荧光双标记示SSeCKS与星形胶质细胞有部分共定位,与神经元无明显共定位。结论:SSeCKS在大鼠脑发育过程中存在时空变化,并且主要定位于星形胶质细胞。提示SSeCKS可能通过参与细胞骨架的调节和信号通路的活化影响大鼠胚胎发育过程中星形胶质细胞指导的神经元的迁移和轴突的生长。
Objective:To investigate the expression of Src-suppressed C kinase substrate,(SSeCKS) in rat brain during its development and its significance.Methods:Western blot was used to test the changes of SSeCKS,and immunohistochemical staining was carried out to investigate its location.Results:SSeCKS was high in the embryonic stage and especially high in the 1-week-old rat brain,while in the 4-week-old rat brain,it was in its lowest level.Regional-distribution analysis showed that expression pattern of SSeCKS in telencephalon,hippocampus and diencephalon was in accordance with above results.Immunohistochemistry results suggested that SSeCKS was in diffuse and granular-like distribution.Part of GFAP positively stained astrocytes also showed SSeCKS immunoreactivity and no striking colocalization was observed between SSeCKS and NeuN.Conclusions:Our results provide evidence that SSeCKS has a temporospatial change during rat brain development and are mainly located in astrocytes,which suggests that SSeCKS may be involved in the process of astrocytes guiding migration and axonal growth of neurons during embryonic development.
