摘要
目的:观察肝癌发生发展过程中血管生成素-2(Ang-2)的动态改变及其与血管内皮生长因子(VEGF)表达的关系。方法:以2-乙酰氨基芴诱发雄性SD大鼠肝细胞癌变,观察癌变过程中肝组织的病理学变化,以免疫组织化学法或酶联免疫吸附法(ELISA)观察Ang-2和VEGF动态表达及相互关系。结果:诱癌过程中鼠肝细胞出现颗粒样变性、不典型增生、癌前病变及肝癌形成的动态变化;血Ang-2水平,在正常对照组为105.23±19.23ng/L、肝细胞变性组为140.17±25.16ng/L、癌前病变组为203.75±23.83ng/L和癌变组为222.39±23.31ng/L,随着病理组织学的改变呈梯度增加,肝癌组明显高于对照组和变性组(P<0.001);癌变过程中Ang-2与VEGF的动态改变呈显著正相关(r=0.785,t=8.00,P=0.000)。结论:肝细胞癌变过程中Ang-2及VEGF呈过表达状态,肝癌的发生发展与新血管生成密切相关。
Objective:To investigate the dynamic expression of hepatic angiopoietin-2(Ang-2) and association with vascular endothelial growth factor(VEGF) during the occurrence and development of hepatocellular carcinoma(HCC).Methods:The dynamic changes of rat liver pathology were observed through the hepatoma model.The expression of circulating Ang-2 and VEGF was analyzed by ELISA or immohistochemistry and their relationship was explored.Results:The hepatic Ang-2 and VEGF expression inereased progressively during the HCC development course from normal hepatocytes to the denatured hepatocytes to precancerous to the formation of HCC.The levels of circulating Ang-2 expression were 105.23±19.23ng/L in the control group,140.17±25.16ng/L in the denatured group,203.75±23.83ng/L in the precancerous group,and 222.39±23.31ng/L in the cancerous group,respectively.There were significant differences between the control group and any of the experimental groups(P〈0.001) and closely positive relationship between Ang-2 and VEGF(r=0.785,t=8.00,P〈0.001).Conclusions:The expressions of hepatic Ang-2 and VEGF are overexpressed,and the occurrence and development of HCC are associated with new angiogenesis.
