摘要
以苏州三山岛上野生水仙鳞茎为材料,通过4℃低温预处理4~10周与未经过低温预处理(室温)对照,再以双鳞片为外植体,通过5种激素(6-BA、NAA、KT、2,4-D、IBA)的配比组合试验,筛选出苏州野生水仙分化最适培养基,建立快繁体系。结果表明:未经过低温预处理的鳞茎切片污染较重,成活率最低为5.0%,最高仅25.0%。经过4℃低温预处理4~10周的鳞茎切片污染率显著降低,成活率最低为25.0%,最高达91.7%;诱导的最佳培养基为MS+6-BA3.2mg/L+NAA0.02mg/L,平均诱导产生小鳞茎4~5个,增殖率达322.22%。
The wild Narcissusin Three-Mount Island of Soochow were respectively dealt with room temperature and low temperature in 4℃during 4-10weeks.The double scales were used as explants and inoculated on 5kinds of different hormone to screen the best medium.The results showed that the lowest survival rate of bulbs in room temperature was 5.0%,and the highest was only 25.0%.Another at 4℃during 4-10weeks was 25.0%,and the highest was up to 91.7%.MS+6-BA 3.2mg/L+NAA 0.02mg/L had the best effect on the inducement,the average of inducing small bulbs was 4-5and the increment rate was up to 322.22%.
