黄芪甲苷对体外炎症状态下单核细胞NF-κB及糖皮质激素受体表达的影响 被引量:20

The Effect of Astragaloside IV on NF-κB and Glucocorticoid Receptor in Monocytes in External Inflammation State

摘要 目的:研究体外炎症状态下,黄芪甲苷对单核细胞糖皮质激素受体和NF-κB及主要细胞因子表达水平的影响。方法:以脂多糖(LPS)刺激体外培养的大鼠单个核细胞以建立体外炎症模型;按照黄芪甲苷95%有效量(ED95)干预炎症状态下的单核细胞,实验分为6组:①正常单核细胞对照组(NC);②黄芪甲苷对照组(AS);③脂多糖对照组(LPS);④糖皮质激素实验组(LPS+Dex);⑤黄芪甲苷实验组Ⅰ(LPS+As);⑥黄芪甲苷实验组Ⅱ(LPS+As+Dex)。用反转录PCR(RT-PCR)和Western blot方法检测糖皮质激素受体(GR)、核因子κB(NF-κB)和肿瘤坏死因子α(TNF-α)表达水平。结果:①LPS刺激后GRαmRNA表达量有所下降,加激素组与单纯LPS刺激比较无明显差异,黄芪甲苷可使GRαmRNA表达量表达显著增加;②LPS刺激后GRβmRNA表达显著增加,加激素或黄芪甲苷后与单纯LPS刺激比较均无明显差异;③LPS刺激后NF-κBmRNA表达显著增加,加激素或黄芪甲苷后与单纯LPS刺激比较均无明显差异;④LPS刺激后TNF-αmRNA表达增加,地塞米松能抑制其表达,单用黄芪甲苷对其表达也具有抑制作用,联合激素和黄芪甲苷对其抑制作用更明显。结论:黄芪甲苷能显著增加GRα的表达,与地塞米松联合能显著抑制细胞因子TNF-α的表达。 Objective:To investigate the effect of Astragaloside IV on NF-κB and glucocorticoid receptor and cytokines in monocytes in external inflammation state.Methods: Lipopolysaccharide(LPS) was introduced in medium of Mono-nuclear cells to come into being inflammation model in vitro.Then the cells were divided into six groups.The method of reverse transcription-polymerase chain reaction(RT-PCR) and Western blot were introduced in this experiment to detect the expression of glucocorticoid receptor,tumor nacrosis factor and nuclear factor-KB.Results: ①LPS reduce the expressions of Glucocorticoid receptor α and Astragaloside improve the expressions of Glucocorticoid receptor α;②LPS improve the expressions of Glucocorticoid receptor β,Astragaloside have no obvious impact on the expressions of Glucocorticoid receptor β;③LPS improve the expressions of NF-κB,Astragaloside have no obvious impact on the expressions of NF-κB;④LPS improve the expressions of TNF-α,Astragaloside reduced the expression of TNF-α.Conclusions: The protective effect of Astragaloside IV may be related to improving the expressions of GRα and inhibited the expression of inflammatory cytokines.
