足阳明经穴特异性的代谢组学模式识别研究 被引量:14

摘要 目的采用基于核磁共振(1H nuclear magnetic resonance,NMR)的代谢组学方法分别观察针刺阳明经穴与他经穴位对健康男性血浆代谢产物的影响,运用模式识别方法分析阳明经穴是否具有共性,与他经穴位是否具有差异性。方法将60例健康青年男性随机分为足三里组、梁门组、巨髎组、阳陵泉组、委中组和空白对照组,每组10例。分别针刺相应穴位,连续针刺5 d,抽取所有受试者的空腹静脉血,采用1H NMR检测血浆代谢物,运用主成分分析和正交偏最小二乘判别分析等模式识别方法分析所得数据。结果 1H-NMR和模式识别相结合的代谢组学方法能较好地区分出足三里组、阳陵泉组、委中组,但对足三里组、梁门组和巨髎组的区分并不明显。针刺阳明经穴位主要影响血浆中的小分子代谢物,与能量代谢通路密切相关;针刺阳陵泉主要影响血浆代谢物中的大分子物质,与脂代谢、脂转运密切相关;针刺委中对血浆代谢物影响不大。结论研究结果提示,阳明经穴具有一定的共性,与阳陵泉、委中均不同;基于NMR和模式识别的代谢组学技术为从代谢角度区分阳明经穴和他经穴位提供了实验证据,有可能成为研究经穴特异性的新的有用工具。 Objective To investigate the effects of acupuncture at Yangming meridian points and other meridian points on plasma metabolites in healthy males using metabonomics based on 1H nuclear magnetic resonance(NMR) and analyze by pattern recognition if Yangming meridian points have common characteristics and if they have differences from other meridian points.Methods Sixty healthy young males were randomly allocated to Zusanli,Liangmen,Juliao,Yanglingquan,Weizhong and blank control groups,10 persons each.The corresponding points were separately acupunctured for 5 consecutive days.Fasting venous blood was taken in all the subjects.Plasma metabolites were measured by 1H NMR.The obtained data underwent principal component analysis and orthogonal partial least squares discriminant analysis used as pattern recognition methods.Results Metabonomics combining 1H NMR and pattern recognition could better discriminate among the Zusanli,Yanglingquan and Weizhong groups but could not well do among the Zusanli,Liangmen and Juliao groups.Acupuncture at Yangming meridian points influenced mainly plasma micromolecular metabolites and was closely related to energy metabolism pathway.Acupuncture at Yanglingquan influenced mainly plasma macromolecular metabolites and was closely related to lipid metabolism and transport.Acupuncture at Weizhong did not largely influenced plasma metabolites.Conclusion This study suggests that Yangming meridian points have certain characteristics,which are different from those of both Yanglingquan and Weizhong.Metabonomics techniques based on NMR and pattern recognition provide experimental evidence for distinguishing between Yangming meridian points and other meridian points from the metabolic aspect and may become a new useful means to study the specificities of meridian points.
