摘要
目的探讨神经细胞相关标志物β-tubulin、NF及GFAP在新生儿外周血的表达情况,为新生儿缺氧缺血性脑病(HIE)治疗中干细胞的动员、分化提供理论依据。方法用Ficoll hypaque分离液分离新生儿外周血,收集单核细胞层,接种于6孔板。37℃饱和湿度的CO2孵箱培养,7 d后更换培养液,弃去未贴壁细胞,贴壁细胞用免疫细胞化进行细胞性质鉴定。结果细胞表达神经元特异性标志(β-tubulin及NF)和星形胶质细胞特异性标志GFAP。结论神经细胞标志物β-tubulin、NF及GFAP在新生儿外周血中表达阳性,新生儿外周血可能存在神经干细胞,这可能是缺氧缺血性脑病自我修复原因之一。
Objective Stem cell-based therapy has the potential to rescue and replace the ischemic tissue caused by hypoxic-ischemia in murine hypoxic-ishcemic encephalopathy(HIE) model.To evaluate the expression of neural-specific molecules,such as neurofilament(NF),βⅢ tubulin and glial fibrillary acid protein(GFAP),in peripheral blood mononuclear cells of neonates,thus providing the theoretic evidence for the stem cells mobilization and differentiation in HIE treatment. Methods Mononuclear cells(MNCs) from neonatal peripheral blood were collected,isolated by centrifugation over lymphoprep(density 1.077/mL) density gradient and then planted in plastic culture flask.Immunocytochemistry was used for cell identification. Results Neurofilament(NF),βⅢ tubulin and glial fibrillary acid protein(GFAP) were detected on isolated mononuclear cells derived from neonatal peripheral blood. Conclusion Neonatal peripheral blood mononuclear cells express some neural-specific molecules,such as neurofilament(NF),βⅢ tubulin and glial fibrillary acid protein(GFAP),providing evidence of existence of neural stem cells in neonatal peripheral blood.This may account for recovery of mild HIE.
