黄芩单细胞克隆愈伤组织生物量积累变化曲线的建立与分析 被引量:2

摘要 [目的]筛选黄芩单细胞克隆优良品系。[方法]选取第10代黄芩单细胞克隆(HQSC)6个品系的愈伤组织作为试验材料,并以黄芩根亲本细胞愈伤组织(0号)为对照(CK),继代后比较不同样本的净增长量。[结果]1~30d培养期内,HQSC各品系愈伤组织生物量积累均呈"S"形曲线变化,其中7、7S号愈伤组织生物量积累速度较快;HQSC各品系愈伤组织延迟增长期为培养后5~8d,对数增长期为培养后9~23d,稳定期为培养后24~30d;在对数增长期内,7S号HQSC愈伤组织的增长量最大,1号HQSC愈伤组织的增长量最小;在稳定增长期内,7号HQSC愈伤组织的增长量最大,1号HQSC愈伤组织的增长量最小。[结论]7、7S号HQSC愈伤组织的增长速度较快。 [Objective] The research aimed to screen out good single-cell cloned line of Scutellaria.[Method] The calli of 6 single-cell cloned lines(HQSC)in the 10th generation of Scutellaria were selected as experimental materials,with calli(No.0)of parent cells from Scutellaria root as CK,the net increments of different samples were compared after subculture.[Result] In the culture period of 1-30 d,the biomass accumulations in calli of various HQSC lines varied in "S"-shape curve,among them the biomass accumulation in calli 7 and 7S was faster.The delayed increase period of calli of various HQSC lines was 5-8 d after culture,their increased logarithmic phase was 9-23 d after culture and their stable phase was 24-30 d after culture.In increased logarithmic phase,the increment of HQSC callus 7S was most and that of HQSC callus 1 was least.In stable increase phase,the increment of HQSC callus 7 was most and that of HQSC callus 1 was least.[Conclusion] The increase speed of HQSC calli 7 and 7S was higher.
