RU 486对体外小鼠着床前胚胎的影响

摘要 本文研究了RU486对体外着床前不同时期小鼠胚胎的RNA聚合酶活性及糖蛋白合成的影响。RNA聚合酶活性以~3H-尿嘧啶核苷(~3H-尿苷)的掺入率为指标,糖蛋白合成以~3H-氨基葡萄糖的掺入量为依据。实验发现:10^(-7)M~10^(-5)MRU486使各期胚胎~3H-尿苷及~3H-氨基葡萄糖掺入降低(P<0.05~0.01);在10^(-6)MRU486处理同时加入10^(-7)M~10^(-5)M孕酮,则各期胚胎两种前体的掺入有不同程度的恢复(P<0.05)。实验结果说明RU486可直接作用于着床前小鼠胚胎,而且这种作用与孕酮相拮抗,为RU486抗早孕提供了一种理论依据。 The effect of RU 486 on preimplanted mouse embryo were studied. The incorporationrate of ~3H-uridine and ~3H-glucosamine as marker of RNA polymerase activity and glyco-protein synthesis in different development stages of embryos were measured. Addition of10^(-5)M~10^(-8)M RU 486 reduced the incorporation of both ~3H-uridine and ~3H-glucosamineinto the embryos. The inhibitory effect of RU 486 on the incoporation of the two precu-sors were antagonized by addition of 10^(-7)M~10^(-5)M progesterone. The results demonstratedthat RU 486 could act directly on the mouse embryos during preimplantation stage andthis effect would be inhibited by progesterone.
