摘要
目的研究维吾尔族代谢综合征(MS)患者chemerin在腹部大网膜、皮下脂肪组织的基因表达及其血清蛋白水平的变化,并探讨其变化在MS中的可能作用。方法选取120例慢性胆囊炎和非胆囊炎维吾尔族患者,在排除了慢性胆囊炎对临床生化指标和MS可能的影响后,选择81例慢性胆囊炎择期手术患者作为chemerin研究的对象,并将其分为MS组(39例)和非MS组(42例)。用realtimeRT-PCR方法检测两组脂肪组织chemerinmRNA的表达水平,ELISA法测血浆chemerin浓度,并测量腰围、臀围、舒张压、收缩压、空腹血糖和血脂等。结果①MS组血浆chemerin水平显著高于非MS组(P<0.01);大网膜和皮下脂肪组织chemerinmRNA的表达水平差异在两组之间均无统计学意义。②血浆chemerin水平与皮下脂肪组织chemerinmRNA表达量呈正相关(r=0.469,P=0.001),与年龄、三酰甘油、腰臀围比及胰岛素抵抗指数呈正相关(r=0.434,P<0.01;r=0.368,P=0.003;r=0.251,P=0.031;r=0.225,P=0.041)。结论维吾尔族MS患者血浆chemerin水平显著升高,且与MS的几个关键指标相关,可能在维吾尔人MS的发生发展中起作用。
Objective To investigate the changes of chemerin gene expressions in abdominal subcutaneous and omental adipose tissues,and of chemerin serum protein level,as well as to explore the role of chemerin in metabolic syndrome development.Methods Among 120 Uygur patients with chronic cholecystitis and non-cholecystitis,after ruling out possible influences from chronic cholecystitis on clinical biochemical parameters and metabolic syndrome 81 patients waiting for selective operations were divided into non-(matabolic syndrome,MS) control group (n=42) and MS group (n=39) for our chemerin study.The chemerin mRNA expression levels were measured by real time RT-PCR in abdominal subcutaneous and omental adipose tissues of 42 non-MS controls and 39 subjects.Serum chemerin was measured by ELISA.We measured waist circumferenc,hip circumference,systolic blood pressure,diastolic blood pressure,as well as fasting plasma glucose and blood lipid.Results ①The plasma chemerin in MS subjects was significantly higher than that in non-MS control group(P0.01).There was not significant difference in chemerin gene expressions in abdominal subcutaneous and omental adipose tissues between MS group and non-MS control group.②The level of plasma chemerin was positively correlated with the expression level of chemerin mRNA in subcutaneous adipose tissue(r=0.469,P=0.001),and with age,triacylglycerol,waist hip ratio and homeostasis model assessment of insulin resistance(r=0.434,P0.01;r=0.368,P=0.003;r=0.251,P=0.031;r=0.225,P=0.041).Conclusion Significantly increased plasma chemerin,alnog with its association with key indicators of MS,can be partly responsible for the development of metabolic syndrome in Uygur population.
