摘要
目的观察氯沙坦对自发性高血压大鼠肾脏结缔组织生长因子表达的调控。方法 14周龄雄性自发性高血压大鼠(SHR)随机分为高血压组和氯沙坦组,各8只,14周龄雄性Wistar-kyoto大鼠8只为正常对照组(WKY组),氯沙坦组给予氯沙坦30mg/(kg.d),16周后,观察各组大鼠体质量、收缩压,免疫组化和RT-PCR检测肾脏结缔组织生长因子(CTGF)、Ⅲ型胶原的表达。结果①高血压组肾脏CTGF、Ⅲ型胶原表达明显高于WKY组(P<0.05);氯沙坦组肾脏CTGF、Ⅲ型胶原表达较高血压组明显降低(P<0.05);②CTGFmRNA和蛋白表达量与Ⅲ型胶原mRNA和蛋白表达量显正相关(r1=0.522,r2=0.603,均P<0.01)。结论氯沙坦可能通过抑制SHR肾脏结缔组织生长因子表达,对早期高血压肾间质损害有保护作用。
Objective To observe the effecst of losartan on connective tissue growth factor (CTGF) in kidneys of spontaneously hypertensive rats(SHR).Methods Sixteen 14 week-old male SHR were randomly divided into hypertensive group (SHR group,n=8) and losartan group (T-SHR group,n=8),and eight 14 week-old Wistar-Kyoto male rats (WKY group) were used as normal controls.The subjects of T-SHR group were fed with losartan 30 mg/(kg·d) every morning.After 16 weeks,we observed their body mass and systolic blood pressure,and detected the expressions of CTGF and type Ⅲ collagen in their kidneys by immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR).Results ①The expressions of CTGF and type Ⅲ collagen in the kidneys of SHR group were significantly higher than those in WKY group (P0.05),while the expressions of CTGF and type Ⅲ collagen in the kidneys of T-SHR group was significantly lower than those in SHR group(P0.05).②The expressions of CTGF and type Ⅲ collagen mRNA and protein were significantly correlated(r1=0.522,r2=0.603,P0.01).Conclusion Losartan can alleviate early hypertension-induced renal tubulointerstitial lesion in SHR by inhibiting the expression of CTGF in their kidneys.
