摘要
从猪卵巢中分离卵子提取透明带(ZP)免疫动物,实验研究证明多克隆ZP抗体,对卵巢功能有一定影响。我们制备的三株猪ZP单克隆抗体,它们与人体主要器官无交叉反应。用免疫荧光、ABC免疫组织化学及体外阻断人精卵结合等方法,进行研究。发现有一株单抗(LPD_8)能明显阻断体外人精卵结合。此外有二株单抗(LPD_8,LPC_4)对卵透明带及卵细胞质有交叉反应,而对卵泡颗粒层,卵泡膜,间质细胞等均无交叉反应。这说明卵细胞质内有与上述单抗结合的抗原决定簇,因此透明带与卵细胞质有共同的抗原成分,这一现象支持了ZP由卵细胞产生的理论。本研究结果提示ZP抗体对卵巢功能的影响是由于抗体和卵细胞结合,早期抑制了卵泡的发育而导致卵泡数目减少,内分泌功能下降,而不是直接作用颗粒层和卵泡膜而使激素分泌紊乱。
Three hybridoma cell lines producing monoclonal antibodies (McAb) against porcinezona pellucida have been established (LPD_8, LPD_2, LPC_4). The tissue specificity and crossreaction of two McAb(LPD_8, LPC_4) have been examined against various human tissues usingBA-ELISA. There was no binding with human tissues (heart, liver, spleen, lung, kidney,stomach, jejunum, brain and striated muscles). By the application of immunohistochemicaltechniques, immunofluorescent staining and sperm binding assay. one McAb(LPD_8) has beenfound to possess the ability in blocking human sperm-ovum interaction in vitro. Two MeAb(LPD_8,LPC_4)cross reacted with zona pellucida and cytoplasm of human oocytes but notcross reacted with membrana granulosa. theca folliculi and other tissues. The results indicatethat certain common antigenic determinants present in the cytoplasm of oocyte and zona pel-lucida, and further supported that the zona pellucida might be derived from oocytes. Basedon the characteristics of monoclonal antibodies, the mechanism of the McAb influencingupon tbe ovary have been discussed.
