嗜肺军团菌血清1型上海环境分离株的脂肪酸成分分析及其应用

摘要 目的分析上海不同地区和场所分离的嗜肺军团菌血清1型(LP1型)的菌体脂肪酸组成成分,并探讨利用脂肪酸组成分析对实验菌株进行鉴定和分型的可行性。方法应用气相色谱技术对36株LP1型上海环境分离株进行菌体脂肪酸成分分析,并利用MIDI公司的Sherlock软件系统对实验菌株进行鉴定和聚类分析。结果 36株LP1型上海环境分离株均被鉴定为嗜肺军团菌嗜肺亚种,其主要脂肪酸成分为异构16∶0酸、反异构15∶0酸、16∶0酸、2-羟基异构15∶0酸和w7c 16∶1酸。聚类分析结果为所有上海环境分离株的欧氏距离<10。结论气相色谱法简便、快速,结合Sherlock软件系统可用于嗜肺军团菌的脂肪酸检测分析和鉴定。不同地区和场所分离的各实验菌株的脂肪酸组成无明显区别。 Objective To analyze the cellular fatty acid composition of Legionella pneumophila serotype 1(LP1) strains isolated from the environment in Shanghai,and study the possibility of applying cellular fatty acid composition analysis to LP1 identifying and typing.Methods 36 LP1 strains isolated from the environment in Shanghai were collected and detected for cellular fatty acid composition analysis by gas chromatography.The identification and clustering of the strains were analyzed by MIDI Sherlock software system.Results All strains were detected and identified as Legionella pneumophila subsp.pneumophila,and the major cellular fatty acid compositions of these strains were iso 16∶0,anteiso 15∶0,16∶0,iso 15∶0 2OH and 16∶1 w7c.According to the clustering analysis result,the Euclidian Distance of the environmental strains was 10.Conclusions Gas chromatography as a simple and rapid method can be used to detect the cellular fatty acid composition of Legionella pneumophila and to identify the species with MIDI Sherlock software system.All experimental strains reveal a high homogeneity.
