摘要
采用微波提取法对两种海带样品多糖成分进行提取,通过DEAE-纤维素离子交换柱分离法纯化。结果表明:柱分离时,洗脱流速保持在1.0~1.5mL/min之间,洗脱液为0.04mol/L、0.05mol/L、0.06mol/LNaOH时,两种海带多糖可分别分离纯化出两种硫酸多糖:A1、A2和J1、J2。用硫酸钡浊度法测得硫酸根的含量分别为:6.26%、3.69%、1.02%、3.09%。
For two laminaria samples, polysaccharide was extracted by microwave-assiated extraction method, and purificated by DEAE-cellulose ion exchange column. The results indicated that distilling speed was 1.0-1 .5 mL/min, when the distill solid was 0.04 mol/L, 0.05 mol/L, 0.06 mol/L NaOH, two polysaceharides sulfates in these two sample could be purificated and separetely as: A1, A2, J1, J2. The content of sulfate ion was determinated by barium sulfate turbidimetry method, the result was 6.26 %, 3.69 %, 1.02 %, 3.09 %.
