摘要
目的建立刺五加中刺五加苷E含量的测定方法,并比较不同产地的刺五加中刺五加苷E含量,为进一步研究不同品系的刺五加中刺五加苷E的含量与其遗传学的关系奠定基础。方法刺五加苷E的提取采用超声提取法;选用YMCODS色谱柱(150 mm×4.6 mm,5μm),检测波长220 nm,以0.1%磷酸水溶液-乙腈(89∶11)为流动相,体积流量1.0ml/min。结果超声波提取法对刺五加苷E提取率稍低于溶剂提取法,但操作简便,耗时较短。刺五加苷E的线性范围为0.625~10.004μg(r=0.999 92);平均回收率为100.64%,RSD为2.97%。结论建立的刺五加苷E提取和测定方法适合于快速准确地分析大量样品。
Objective To establish a method for determination of Eleutheroside E content in Eleutherococcus senticosus from different provenances,which will afford a base for further research on the relationship between the heritage of Eleutherococcus senticosus and the content of Eleutheroside E.MethodsA supersonic extraction method was adopted for the extraction of Eleutheroside E.HPLC determination conditions were as follows:YMC ODS column(150 mm×4.6 mm,5 μm),water containing 0.1% phosphoric acid and acetonitrile(89∶11) as mobile phase,with a flow rate of 1.0 ml/min,and UV detect wavelength 220 nm.ResultsAs compared with organic solvent extraction method,supersonic extracting method was easy and time-saving,despite its slightly low extracting rate.The linear range of Eleutheroside E was 0.625-10.004 μg(r = 0.999 92),average recovery coefficient was 100.64%,RSD = 2.97%.ConclusionThe determination method is suitable for the precisely analysis of Eleutheroside E.
