Objectives \ To examine whether the
antiproteinnuric effects of Ttype and Ltype calcium channel antagonists in
streptozotocin(STZ)induced diabetic rat is related to the ameliraation of glucose and lipid
metabolism or not.\ Methods \ Two weeks after being uninephrectomized (UNx), male SD rats
were insulted to have diabetes by STZ. Ttype calcium channel antagonist mibefradil (30
mg/kg),Ltype calcium channel antagonist amlodipine (30 mg/kg), angiotensin converting
enzymeinhibitor(ACEI) cilazapril(10 mg/kg) were given respectively by daily gavage for 4 weeks
from the next day of STZ injection. Another two groups, untreated uninephrectomized diabetic
group and uninephrectomized group were served as Author control. Blood pressure was
measured weekly by tailcuff method. 24 h urine was collected in a metabolic cage to determine
proteinuria and creatinine clearance rate(Ccr). Plasma glucose, triglyceride and cholesterol
concentration as well as plasma ACE activity, serum insulin and Cpeptide, HbA1c were
measured.\ Results \ All diabetic rats showed overt proteinuria, increased Ccr, glucoselipid,
insulin, Cpeptide and HbA1c. Treated animals presented less urinary protein and less elevated
Ccr. Although cilazapril group had some decreased in blood glucose and serum cholesterone,
no any changes were detected in mibefradil and amlodipine treated groups. No remarkable
differences in plasma insulin, Cpeptide and HbA1c levels were found among nontreated and
treated diabetes.\ ConclusionBZ]\ The protective effect of calcium channel blockage on diabetic
nephropathy might not mediated by improvement of glucose and lipid metabolism.
