促凋亡基因bax在骨巨细胞瘤中的表达及分化调控意义

Expression and Significance of Proapotosis Gene in Giant cell Tumor of Bone

目的：研究促凋亡基因ｂａｘ在骨巨细胞瘤中的表达及其与Ｊａｆｅ分级和预后的关系。方法：采用兔抗人Ｂａｘ多克隆抗体及免疫组织化学ＳＡＢＣ法。结果：７３例骨巨细胞瘤中，Ｂａｘ阳性４７例（６４４％）；其中Ｊａｆｅ病理分级中Ⅰ级２７例中，阳性２２例（８１５％），Ⅱ级３８例中阳性２３例（６０５％），Ⅲ级８例中，２例阳性（２５０％），Ｐ＜００５。在不同预后分组中Ｂａｘ阳性率间无显著差异（Ｐ＞００５）。结论：促凋亡基因在骨巨细胞瘤中不同程度的表达可能参与骨巨细胞瘤凋亡的调节，并与肿瘤分化程度有关。单一Ｂａｘ表达不能反应预后。

Objective:To study the expression of proapoptosis gene Bax in giant cell tumor of bone,and its relationship with Jaffe pathologic grading and prognostic significance.Method:Streptavidin Biotin peroxidase Complex(SABC) methods,polyclonal antibody against Bax protein on archival material were employed.Results:Bax was positive in 47 cases of all 73 specimen of giant cell tumor.According to Jaffe pathologic grading,81 2%(22/27) of grade Ⅰ specimen were positive;60 5%(23/38) of grape Ⅱ positive;25 0%(2/8) of grade Ⅲ positive.There was significant difference between expression of Bax in grade Ⅰ、Ⅱ、Ⅲ group ( P <0 05).While there was no obvious difference between expression of in cure、recurrence and metastases group.( P >0 05).Conclusion:The result suggests that proapoptosis gene Bax are differently expressed in most of giant cell tumor of bone and Bax may take part in the modulation of apoptosis and is associated with the differentiation of giant cell tumor of bone.Bax is not associated with prognosis.
