摘要
目的:观察三种不同负荷条件下血管内皮生长因子(VEGF)在小鼠心肌组织中的表达,并进一步阐明不同负荷对机体心脏中心室肌和心房肌不同代偿能力的影响规律。方法:雄性2月龄昆明小鼠30只,每只体重(34±1.87)g,用国家标准啮齿类动物饲料饲养,自由进食进水。适应性喂养1周后,随机分为3组,每组10只,对训练组进行训练。训练完成后取其心脏,固定包埋后作HE和免疫组化染色,观察各组的体重改变、VEGF表达状况。利用IMAGE PRO PLUS5.1图像分析软件测量面积比。结果:三组小鼠中120 min+负重组随着训练时间的延长而逐渐不能顺利地完成任务,游完后都表现筋疲力尽;而其它两组的小鼠活动状态一直较好。静养对照组VEGF表达为阴性;120 min组表达为弱阳性;120min+负重组表达为阳性。分析其阳性面积比(阳性面积/视野面积),120 min组和120 min+负重组均与静养对照组有明显性差异,120 min组与120 min+负重组也有明显性的差异。结论:一定的运动可以使心肌分泌VEGF,在适宜的时间内,运动的强度越大,VEGF的表达就越强烈,这对心房肌的影响较大,而对心室肌的影响不大,并有一定代偿作用。
Objective:To observe the changes of VEGF expression in mice myocardium under three different load conditions.And further to expound the rules that the regeneration blood vessel ability influenced by the different loads.Methods:As experimental objects,30 Kunming male mice that each one weight(34 ± 1.87)g with feeding by the state standards of rodents feeding and free eating influent were divided into 3 groups randomly after a week feeding adaptability,that was,the control group、the 120 minutes group、the 120 minutes + burdened group.Upon completion of the training,the heart was removed.They were fixed by H.E.and stained immunochemically after embedding.The change in the body weight and VEGF expression were observed.IMAGE PRO PLUS 5.1 image analysis software was used to measure area ratio.Results:120 minutes + burdened group of three groups with the reorganization of the training time could not be extended gradually to the smooth completion of tasks.They had shown exhaustion after training.And the other two groups of mice activities in the state had been better.Three groups of mice in eight weeks of training time drank water and ate better.Their weight had been showing an upward trend.The control group displayed for the negative VEGF expression.The 120 minutes group displayed weakly positive expression.Analysis of the positive area ratio(positive area / field area),120 min and 120min + negative re-group with the aim of keeping the control group were significantly different,120 min and 120 min + group there was a marked reorganization of the negative difference.Conclusions:Certain movement can secrete VEGF.In the appropriate time,with campaign greater,the intensity of VEGF express stronger.And it is not likely to cause serious cardiac injury.Training of athletes at the daily exercise of general population must achieve remarkable results with maintaining a certain degree of effective time.
