摘要
目的探讨缺氧诱导因子-1α(HIF-1α)和嗜酸粒细胞趋化因子(eotaxin)在鼻息肉组织中的表达及其与发病的关系。方法选取45例经鼻内镜手术治疗的鼻息肉患者的鼻息肉组织为实验组,12例行下鼻甲部分切除术治疗的慢性肥厚性鼻炎患者的下鼻甲黏膜为对照组。分别采用免疫组化法和苏木素-伊红染色法检测两组标本中HIF-1α、eotaxin阳性染色细胞数和嗜酸粒细胞数,采用反转录-聚合酶链反应结合半定量分析方法检测HIF-1α、eotaxin mRNA表达水平。结果实验组HIF-1α、eotaxin阳性染色细胞数及嗜酸粒细胞数较对照组明显升高,差异均有统计学意义(P<0.05)。实验组HIF-1α阳性染色细胞数及eotaxin阳性染色细胞数与嗜酸粒细胞数均呈正相关(r值分别为0.545和0.608,P<0.05);实验组HIF-1α阳性染色细胞数与eotaxin阳性染色细胞数呈正相关(r=0.643,P<0.05)。实验组HIF-1α、eotaxin mRNA表达水平较对照组明显升高,差异均有统计学意义(P<0.05)。结论 HIF-1α、eotaxin均可引起鼻息肉中嗜酸粒细胞的浸润,且二者相互促进,对鼻息肉的发病起非常重要的作用。
Objective To investigate the expressions of hypoxia inducible factor-1α(HIF-1α) and eotaxin in nasal polyps tissue and their relationships with nasal polyp.Methods The nasal polyp tissues of 45 patients with nasal polyps treated with nasal endoscopy were taken as trial group,and the inferior turbinate mucosa of 12 patients with chronic hypertrophic rhinitis treated with partial inferior turbinectomy were taken as the control group.The numbers of eosinophils (EOS) infiltration and positive expression cells of HIF-1α and eotaxin in the two groups were detected by using immunohistochemistry and hematoxylin-eosin staining,and HIF-1αand eotaxin mRNA expression were assayed by using reverse transcription-PCR (RT-PCR) in combination with semiquantitative analysis.Results The numbers of HIF-1α and eotaxin-positive staining cells and the EOS number in the trial group were significantly higher than those in the control group (P0.05).The counts of HIF-1α positive staining cells and eotaxin-positive staining cells in the trial group were positively correlated with eosinophil number (r=0.545,0.608,P0.05);HIF-1α positive staining cells and eotaxin-positive staining cells in the trial group were positively correlated (r=0.643,P0.05).The expressions of mRNA of HIF-1α and eotaxin in the trial group were significantly higher than those in the control group (P0.05).Conclusion Both HIF-1α and eotaxin can cause EOS infiltration of nasal polyps.And they promote each other,playing an important role in the development of nasal polyp.
