摘要
[目的]研究适合于贺兰山紫蘑菇(Corinarius rufo-olivaceus)基因组DNA的提取方法。[方法]通过采用改良CTAB法Ⅰ、改良CTAB法Ⅱ、SDS法、Doyle-Doyle法和KI法5种基因组DNA提取方法,分别提取贺兰山紫蘑菇的基因组DNA,用DU800核酸蛋白检测仪测定其DNA含量、浓度和纯度,并通过琼脂糖凝胶电泳进行提取结果比较。[结果]5种不同的提取方法均能提取该紫蘑菇的基因组DNA,其中KI法提取的DNA纯度、浓度和得率为最好。[结论]KI法是贺兰山紫蘑菇(C.rufo-olivaceus)DNA提取的适宜方法,该法具有简单、便捷、省时、适合基因组DNA大量提取的特点。
[Objective]The aim was to study the appropriate method for extracting the genomic DNA of C.rufo-olivaceus.[Method]The genomic DNA was extracted by the five different kinds of DNA extraction methods,such as modified CTABⅠmethod,modified CTABⅡmethod,SDS method,Doyle-Doyle method and KI method.The content,concentration and purity of total DNA were detected by DU800 and the results were compared by electrophoresis of agarose gel.[Result]The five different extraction methods were able to extract the genomic DNA of the C.rufo-olivaceus and the KI method could receive the best of purity,concentration and yield of total DNA.[Conclusion]The KI method was the appropriate method for extracting the genomic DNA of C.rufo-olivaceus.It had the features of simple,convenient,time-saving and could be used to extract genomic DNA in large numbers.
