摘要
[目的]PTLF是杨树花发育的重要基因,可抑制杨树开花,解决杨树飞絮散粉污染环境的问题,并降低生殖生长对营养生长的竞争。[方法]利用农杆菌介导的叶盘转化法将含PTLF基因的RNAi(RNAinterference)载体35S-PTLF-IR转化毛白杨。[结果]通过聚合酶链式反应(PCR)检测,初步验证10株阳性转化植株。[结论]对转基因毛白杨和野生型毛白杨组培苗进行高生长分析表明,前者生长速度、株高低于后者。
[Objective]PTLF is an important gene involved in flowering in poplar,in order to repress poplar flowering and solve the environmental problem caused by male and female catkins,and lessen the competition between vegetative growth and reproductive timber growth.[Method] RNA interference vector of PTLF gene was transferred into Populus tomentosa by agrobacterium mediated method.[Result] PCR identification showed ten transgenic plants were positive.[Conclusion]Growth analysis suggested that transgenic plants grew slower and was lower than wild type control.
