血必净注射液对小鼠脓毒症早期肝脏白细胞聚集及黏附分子ICAM-1/VCAM-1表达的影响

Influence of Xuebijing injection on leukocytes aggregation in liver and expression of ICAM-1/VCAM-1 in mice with sepsis at prophase

目的探讨血必净注射液对小鼠脓毒症早期肝脏白细胞聚集及细胞间黏附分子(ICAM-1)和血管细胞黏附分子-1(VCAM-1)表达的影响。方法将实验动物随机分成4组:正常组(n=8)、假术组8(n=8)、脓毒症组(n=8)、脓毒症加血必净组(n=8)。盲肠结扎穿刺法(CLP)建立动物脓毒症模型。假术组给予相同的麻醉和腹壁正中切口,盲肠外置,然后纳入腹腔。各组动物分别于操作后立即给予血必净1.71 g生药/kg或生理盐水尾静脉静推;检测24 h后动物肝脏组织病理学变化,测定肝组织髓过氧化物酶(MPO)、ICAM-1和VCAM-1蛋白的表达。结果脓毒症组24 h肝组织中白细胞自血管内移出增加,MPO活性增强,同时肝组织ICAM-1和VCAM-1蛋白表达显著升高。血必净注射液干预后肝组织内白细胞聚集明显减少,肝组织MPO被抑制(P<0.01),ICAM-1、VCAM-1蛋白表达量均被显著抑制(P均<0.01)。结论血必净注射液能明显抑制肝组织MPO活性,减少肝组织ICAM-1和VCAM-1蛋白的表达水平,从而减轻严重烧伤后组织白细胞聚集,改善肝功能,有效减轻肝脏炎症反应。

Objective It is to approach the influence of Xuebijing injection on leukocytes aggregation in liver and expression of ICAM-1/VCAM-1in Mice with sepsis at prophase.Methods The laboratory animals were randomly divided into normal group,sham operation group,sepsis group and sepsis plus Xuebijing group in which there were eight mice.The sepsis animal models were established with cecum ligation puncture method.The sham operation group was done the same anesthesia and midline laparotomy,the cecum was exteriorized and returned to the abdominal cavity.After operation,the animals in all the groups were immediately given Xuebijing 1.71 g/kg or physiological saline intravenous push to caudal vein respectively.After 24 h,the pathology change of liver tissue was detected and MPO and the expressions of ICAM-1 and VCAM-1 were determined.Results In sepsis group after 24 h,leukocytes aggregation in liver tissue was increased,MPO activity was strengthened and the expressions of ICAM-1 and VCAM-1 in liver tissue were significantly heightened.After Xuebijing injection interfered,leukocytes aggregation in liver tissue was obviously decreased,MPO in liver tissue was inhibited（P〈0.01） and the expressions of ICAM-1 and VCAM-1 were both significantly inhibited（both P〈0.01）.Conclusion Xuebijing injection can obviously inhibit MPO activity in liver tissue and decrease the expression levels of ICAM-1 and VCAM-1 in liver tissue.Thus it can lessen leukocytes aggregation in tissue after severe burn,improve liver function and effectively lessen liver inflammatory reaction.
