山薯组培苗继代培养基配方筛选 被引量:8

Screening the subculture medium for Dioscorea fordii Prain et Burk plantlets

摘要 以山薯组培苗带腋芽茎段为外植体,以MS为基本培养基,探讨植物生长调节剂6-BA(0.5、1.0、2.0 mg/L)或TDZ(0.01、0.10、0.50、1.00、2.00、4.00 mg/L)、不同培养方式和活性炭对其继代培养的影响。结果表明,添加不同浓度6-BA对山薯组培苗增殖系数、茎长及腋芽点芽球的影响不明显,而随着TDZ浓度的升高,山薯组培苗增殖系数和茎长逐渐降低而芽球显著增加,茎段腋芽生长受到明显抑制。山薯组培苗较优的继代培养基为MS+1.0 mg/L 6-BA+0.1 mg/LNAA+30 g/L蔗糖+1.5 g/L活性炭的液体培养基。液体培养基利于促进山薯组培苗增殖,其增殖系数、茎长、植株鲜重和干重显著高于固体培养基。活性炭的添加可以减少褐化,促进组培苗的增殖培养。此外,培养方式与活性炭交互作用对组培苗茎长、增殖系数、植株鲜重及干重无显著影响。 The shoots with axillary buds derived from D.fordii(Guihuai 2) plantlets were used as explants to investigate the effects of different plant growth regulators,viz.,6-BA(0.5,1.0 and 2.0 mg/L and TDZ(0.01,0.10,0.50,1.00,2.00 and 4.00 mg/L)in MS medium,culture media(solid or liquid) and activated carbon(AC) on subculture of plantlets.The results showed that 1.0 mg/L 6-BA supplemented in MS basal medium had no significant effects on proliferation coefficient,stem length and gemmules derived from axillary buds.With the increase in TDZ concentration in MS medium,the proliferation coefficient,stem length of proliferated plantlets and growth of axillary buds inhibited significantly,while number of gemmule increased.The liquid MS medium +1.0 mg/L 6-BA+0.1 mg/L NAA+30.0 g/L Sugar +1.5 g/L AC was found to be the best medium for shoot proliferation.Compared to semi-solid culture,the liquid culture promoted shoot proliferation significantly,proliferation coefficient,stem length and fresh weight and dry weight of subculture plantlets were also found higher.Addition of AC in MS medium reduced the browning of explants and promoted the shoot proliferation.Furthermore,the interaction between culture type and AC in MS medium had no significant effects on the proliferation coefficient,stem length,fresh weight and dry weight of subculture plantlets.
