摘要 目的观察电针对体外培养星形胶质细胞(Astrocyte,Ast)机械性损伤后反应性胶质化的影响。方法取新生3d龄SD大鼠的大脑皮层进行Ast原代培养,取第三、四代细胞制作细胞机械性损伤模型,电针刺激,用倒置相差显微镜及免疫细胞化学荧光染色观察细胞生长增殖,细胞形态及胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达的变化。结果模型组Ast机械性损伤后,细胞增生,体积变大,GFAP表达增强。电针组较模型组细胞增生减少,细胞体积变化不大,GFAP表达较弱。结论电针刺激可以抑制体外培养Ast机械性损伤后的增生,降低细胞内GFAP的表达,即电针可以抑制Ast的反应性胶质化。 Objective To explore the effect of electroacupuncture(EA) on reactive gliosis of astrocytes in vitro after mechanical injury.Methods Neonatal 3d-age SD rats were selected to carry out primary culture of cerebral astrocytes.The mechanical injury model was product using the third or fourth generation cell.The cells growth and proliferation,cell morphology and glial fibrillary acidic protein(GFAP) expression were investigated in EA group and control group.Results After mechanical injury,astrocytes in the control group proliferate with larger size and more GFAP expression.The cell proliferation and GFAP expression decreased,cell volume changed little in EA group.Conclusion Electrical stimulation can reduce the astrocyte proliferation and the expression of GFAP in vitro after mechanical injury,indicating that EA can weaken the response of astrocytes in gliosis.
