摘要
目的调查中国汉族人的节律基因period3的基因型频率。方法采集普通健康汉族人血液样本240份。提取基因组DNA,PCR扩增per3基因的可变数目串联重复(variable-number tandem-repeat,VNTR)序列,鉴定个体的per3基因型。结果确定汉族人中per34等位基因频率为0.883,per35的等位基因频率为0.117;per3基因型频率:PER34/4为78.33%,PER34/5为20.00%;PER35/5为1.67%。与高加索人、非裔美国人和意大利人的per3基因型频率比较差异有统计学意义(χ2=85.61,P<0.01)。结论汉族人的per3基因型频率与高加索人、非裔美国人和意大利人的基因型频率不同。
Objective To investigate the frequency distribution of circadian clock gene period3 in Han Chinese.Methods Blood samples were collected from healthy Han Chinese volunteers.Genomic DNA was extracted from leucocytes.Genotyping was performed using polymerase chain reaction.Results The genotype frequency of Han Chinese was 78.3% for PER3^4/4,20.0% for PER3^4/5,1.7% for PER3^5/5.The frequency distribution of per3 of Han Chinese was significantly different from that of Caucasians,African Americans and Italian(χ^2=85.61,P〈0.01).Conclusion Per3 genotypic and allelic frequencies show significant difference between Han Chinese and Caucasians,African Americans and Italian.
