自噬基因Beclin1在宫颈鳞癌中的表达及其与HPV感染的关系 被引量:5

摘要 目的探讨自噬基因Beclin1在宫颈鳞癌中的蛋白表达以及对人乳头状瘤病毒(HPV)16E6、E7感染的影响。方法采用免疫组化SP法检测81例宫颈鳞癌、20例宫颈上皮内瘤样病变(CIN)Ⅱ-Ⅲ级和20例正常宫颈组织中自噬基因Bec-lin1的表达,采用PCR扩增法检测宫颈鳞癌组织中HPV16E6、E7的DNA状况。结果宫颈鳞癌组织中Beclin1阴性、弱阳性、强阳性表达率依次是43.2%(35/81),34.6%(28/81),22.2%(18/81),分别与正常宫颈组15.0%(3/20)、30.0%(6/20)、55.0%(11/20)和宫颈上皮内瘤样病变组20.0%(4/20)、30.0%(6/20)、50.0%(10/20)相比,Beclin1在宫颈鳞癌中的表达存在显著下调(P=0.009,P=0.033)。HPV16E6、E7在宫颈鳞癌中的阳性率分别为66.7%和58.0%,HPV16E6、E7感染与宫颈鳞癌中Beclin1蛋白的表达无相关性(r=-0.126,P=0.262;r=-0.125,P=0.270)。结论自噬基因Beclin1在宫颈鳞癌中表达下调,其表达对HPV16E6、E7感染无明显影响。 Objective To investigate the expression of autophagy gene Beclin 1 in cervical squamous cell carcinoma and its effect on papillomavirus 16 infection.Methods SP immunohistochemistry was used to detect the expression of Beclin 1 in 81 cases of cervical squamous cell carcinoma,20 cases of cervical intraepithelial neoplasm(CIN)Ⅱ-Ⅲ,and 20 cases of normal cervix.Polymerase chain reaction(PCR)was used to detect the expression of HPV16 E6,E7.Results The rates of negative,weak,strong positive expression of Beclin 1 were 43.2%(35/81),34.6 %(28/81),22.2%(18/81)in cervical squamous cell carcinoma,respectively,15.0%(3/20),30.0%(6/20),55.0%(11/20)in normal cervix and 20.0%(4/20),30.0%(6/20),50.0%(10/20)in CIN Ⅱ-Ⅲ.The strong positive rate of Beclin 1 in cervical squamous cell carcinoma was significantly lower than that in normal cervix(P=0.009)and CIN(P=0.033).The infection rates of HPV16 E6 and E7 were 66.7% and 58% in the invasive carcinoma,respectively.The expression of Beclin 1 was not correlated with the infections of HPV16 E6,E7(rE6=-0.126,P=0.262;rE7=-0.125,P=0.270).Conclusion Expression of autophagy gene Beclin 1 is down-regulated in cervical squamous cell carcinoma,which is not associated with the infection of HPV16 E6,E7.
