细胞内游离Ca^(2+)和cAMP浓度在胃癌细胞凋亡过程中的变化特点

Change of Intracellular-free Calcium and cAMP levels in the Process of Gastric Cancer Cellular Apoptosis

目的：探讨胃癌细胞凋亡过程中细胞内游离Ｃａ２＋（［Ｃａ２＋］ｉ）浓度和环磷酸腺苷（ｃＡＭＰ）浓度的变化特点及意义。方法：选择顺铂诱导胃癌细胞凋亡，采用光镜、电镜、琼脂糖凝胶电泳和流式细胞仪检测凋亡；Ｆｕｒａ－２荧光负荷技术测［Ｃａ２＋］ｉ，竞争性蛋白结合法测ｃＡＭＰ浓度。结果：顺铂浓度为２μｇ／ｍｌ作用于胃癌细胞２４小时出现典型的凋亡改变，即凋亡小体、细胞皱缩，染色质固缩和片段化，ＤＮＡ梯状条带，流式细胞仪上见“凋亡峰”；胃癌细胞凋亡过程中［Ｃａ２＋］ｉ明显升高，以凋亡早期更为明显，ｃＡＭＰ浓度也高于对照组。结论：［Ｃａ２＋］ｉ和ｃＡＭＰ浓度在胃癌细胞凋亡过程中表现为上调趋势。
