大鼠脊髓损伤后MMP-9表达和细胞凋亡的研究 被引量:5

摘要 目的探讨MMP-9在SCI后继发性细胞凋亡中的作用,为继发性脊髓损伤的治疗提供新的思路。方法成年雄性SD大鼠84只,随机分为假手术组与损伤组(spinal cord injury group,SCI group),两组大鼠均于SCI后1 h、6 h、12 h、1 d、3 d、5 d、7 d(每个时间点n=6)处死大鼠取材。免疫组织化学染色观察损伤脊髓组织MMP-9表达并测平均光密度值(optical density,OD),原位缺口末端标记法(TUNEL法)观察损伤脊髓组织细胞凋亡并计算凋亡指数(apoptotic Index,AI)。分别对各时间点损伤脊髓组织MMP-9平均光密度和细胞凋亡指数进行组间、组内比较,并对损伤组各时间点MMP-9平均光密度和细胞凋亡指数进行相关性分析。结果大鼠SCI后损伤节段脊髓组织中MMP-9表达和细胞凋亡指数明显增多,24 h达高峰,持续至3 d后逐渐下降,7 d仍有表达。结论大鼠SCI后损伤节段脊髓组织的MMP-9表达与细胞凋亡呈正相关关系(相关系数r=0.936,P<0.05)。
