瑞香狼毒药材中蛋白质部位的体外活性研究

Study on in vitro Activity of Protein Site from Stellera chamaejasme L.

目的建立瑞香狼毒药材蛋白质部位体外活性研究的方法。方法琼脂糖扩散法测定总蛋白的抑菌活性;MTT细胞培养法测定总蛋白及分离得到的6个分子量蛋白对肝癌细胞生长的抑制作用及对正常细胞的毒性作用。结果体外活性研究结果表明,总蛋白没有抑菌活性但具有抗肿瘤活性和细胞毒性;分离得到的6个分子量的蛋白中2、3、4、6对肿瘤细胞有抑制作用,1、3、4对正常细胞有毒性作用。结论实验方法可作为瑞香狼毒药材中蛋白质生物活性研究的方法,为瑞香狼毒蛋白质部位的毒性蛋白及活性蛋白的研究提供了方法依据。

Objective To set up the method of detecting in vitro activity of protein site from Stellera chamaejasme L.MethodsThe method of agar diffusion was used antibacterial activity of total protein;MTT cell cultural technique was adopted to measure the cell activity of total protein and the six molecular weight proteins isolated from total protein.ResultsThe study of in vitro activity indicated that the total protein was lack of antibacterial activity but had the function of anti-tumor and the activity of toxicity;the proteins of 2,3,4,6 had the function of anti-tumor and the 1,3,4 have the toxic effect to normal cells.Conclusion In this paper,the method can be used to study the biological ammity of the protein of Stellera chamaejasme L.
