良性前列腺增生上皮对间质细胞S100A11基因表达的影响

摘要 目的:研究良性前列腺增生(BPH)上皮对间质细胞S100A11基因表达的影响。方法:分离BPH的间质细胞与上皮细胞,经形态学及免疫学方法证实后,构建共培养模型,并提取单独/共培养条件下间质细胞的mRNA,RT-PCR检测S100A11表达变化。结果:成功分离前列腺间质与上皮细胞并构建共培养模型,S100A11基因mRNA的表达在有上皮细胞共培养的间质细胞中表达低于无上皮细胞共培养的间质细胞,S100A11/G3PDH的灰度比值分别为0.865±0.10、0.963±0.13,差异有统计学意义(P<0.01)。结论:BPH上皮细胞可抑制间质细胞S100A11基因表达。 Objective:To investigate the influence of epithelial cells on the S100A11 expression in stromal cells of benign prostale hyperplasi(BPH).Methods:To separate and culture prostate epithelial cells and stromal cells identified by the cell morphology and immunohistochemistry in the tissue of BPH.After the stromal-epithelial co-cultured model was constructed,the expression of S100A11 mRNA was detected by RT-PCR in stromal cells with or without the epithelial cells co-cultured.Results:A stromal-epithelial co-cultured model was obtained.The expression of S100A11 mRNA was lower in stromal cells with epithelial cells co-cultured than that in stromal cells without epithelial cells co-cultured.The ratio of gray level between S100A11 and G3PDH in stromal cells with or without epithelial cells co-cultured was 0.865±0.10 and 0.963±0.13(P〈0.01).Conclusion:The epithelial cells can suppress the S100A11 expression of stromal cells in BPH.
