摘要
目的:探讨耐紫杉醇卵巢癌细胞株SKOV3/TAXOL中凋亡途径相关基因的表达差异。方法:运用real-ti me-RT-PCR技术,检测耐紫杉醇卵巢癌细胞株SKOV3/TAXOL和敏感细胞株SKOV3中凋亡途径相关基因TRADD、DFFA、MADD和TGM2的表达量,并对结果进行对比分析。结果:4个基因在SKOV3/TAXOL中的表达量均高于SKOV3,其中TRADD、DF-FA、MADD、TGM2分别高2.25、3 333、3.5×105和2 865倍。结论:凋亡相关基因TRADD、DFFA、MADD和TGM2在SK-OV3/TAXOL细胞株中表达都升高,说明卵巢癌细胞对紫杉醇耐药可能在一定程度上与细胞凋亡功能异常相关。
Objective:To study the differential expression of apoptosis related genes such as TRADD,DFFA,MADD and TGM2 in ovarian cancer cell line resistant to paclitaxel SKOV3/TAXOL.Methods:The expression of TRADD,DFFA,MADD and TGM2 in SKOV3/TAXOL and SKOV3 was measured using real time RT-PCR and the results in the two cells were also analyzed and compared.Results:The expression of TRADD,DFFA,MADD and TGM2 in SKOV3/TAXOL was elevated 2.25,3 333,3.5×10^5 and 2 865 times than that in SKOV3,respectively.Conclusion:The expression of TRADD,DFFA,MADD and TGM2 in SKOV3/TAXOL was higher than that in SKOV3.Ovarian cancer cell resistant to paclitaxel may be relatable somewhat with abnormal apoptosis.
