摘要
目的:建立测定洋金花药材中东莨菪碱含量的高效液相色谱法。方法:采用Hypersil(5μm,46mm×200mm)色谱柱,以甲醇-水(53∶47,水中含20mmol·L-1醋酸钠,002%三乙胺,03%四氢呋喃,pH为686)为流动相,紫外检测波长215nm,外标法定量。结果:东莨菪碱浓度在2~666μg·mL-1范围内呈线性关系,方法回收率为9909%(n=6),RSD为074%。结论:此法灵敏度高,重现性好。
Objective: A silica gel high performance liquid chromatographic method was developed for the determination of scopolamine. Methods: Using a Hypersil column (5 m, 46 mm200 mm), the mobile phase consisted of methanol-water (5347, with 002% triethanolamine, 20 m molL-1 sodium acetate and 3% tetrahydrofuran, pH 686); The UV detection wavelength was 215 nm. The concentration of scopolamine was quantified by external standard method. Results: Within 2-666 gmL-1, scopolamine had a good linearity. The average recovery was 9909% with RSD of 074%. Conclusion: The method is sensitive and reproducible.
