高效液相色谱法测定洋金花药材中东莨菪碱的含量

Determination of Scopolamine in Flos Daturae by HPLC

目的：建立测定洋金花药材中东莨菪碱含量的高效液相色谱法。方法：采用Ｈｙｐｅｒｓｉｌ（５μｍ，４６ｍｍ×２００ｍｍ）色谱柱，以甲醇－水（５３∶４７，水中含２０ｍｍｏｌ·Ｌ－１醋酸钠，００２％三乙胺，０３％四氢呋喃，ｐＨ为６８６）为流动相，紫外检测波长２１５ｎｍ，外标法定量。结果：东莨菪碱浓度在２～６６６μｇ·ｍＬ－１范围内呈线性关系，方法回收率为９９０９％（ｎ＝６），ＲＳＤ为０７４％。结论：此法灵敏度高，重现性好。

Objective: A silica gel high performance liquid chromatographic method was developed for the determination of scopolamine. Methods: Using a Hypersil column (5 m, 46 mm200 mm), the mobile phase consisted of methanol-water (5347, with 002% triethanolamine, 20 m molL-1 sodium acetate and 3% tetrahydrofuran, pH 686); The UV detection wavelength was 215 nm. The concentration of scopolamine was quantified by external standard method. Results: Within 2-666 gmL-1, scopolamine had a good linearity. The average recovery was 9909% with RSD of 074%. Conclusion: The method is sensitive and reproducible.
