摘要
以2 年生杜仲带芽茎段为起始材料进行离体快繁时,最佳取材时间是5 月份。在初代培养中,同一枝条上中段腋芽容易萌发,茎段竖插于培养基比横放好,在一定浓度范围内,培养基中细胞分裂素与生长素浓度比为10 :1 时腋芽萌发率最高。将腋芽萌发后长成的新技转入添加 B A P(1 .0 m g/ L) 和 N A A(0 .1 m g/ L) 的培养基中,第二轮增殖效果较好,以后则出现玻璃化等不正常现象,只有在培养基中加入 Ag N O3 后才能使茎芽的生长恢复正常。由快繁产生的枝条经壮苗培养后转入1/4 M S+ 间苯三酚( P G)10 m g/ L+ I B A2 .0 m g/ L 培养基中,生根率较高(43 .0 % ) 。
In propagation through enhanced axillary branching of two-years old Eucommia ulmoides, May proved to be a favorite time for explant collection. During the initial period of culture with these explants, axillary buds on the middle section of a branch developed readily into a shoot when the explants were implantd vertically in the medium, and the ratio between cytokinin and auxin was kept at 10:1. After the newly developed shoots were transfed into medium supplemented with BAP and NAA, multiplication through axillary branching took place normally druing the first passage, however, vitrification and other abnormalities appeared during the following passage of multiplication unless AgNO 3, was added into the medium. Shoots produced in this way were then strenthened and rooting was finally induced upon transfering th estrengthened shoots onto 1/4MS medium supplemented with PG and IBA. The percentage of shoots rooted could be as high as 43%.
