摘要
目的:寻找人肝细胞癌(HCC)特异抗原。方法:应用重组克隆表达抗原的血清学鉴定技术(SEREX),用病人自身血清从广西肝细胞癌cDNA文库中筛选出编码HCC肿瘤抗原的基因克隆,用肝癌患者及其他人血清检验HCC肿瘤抗原对肝癌的特异性,测定抗原DNA序列并与基因库核对寻找其同源结构。结果:其中一种HCC肿瘤抗原与衰老标记蛋白-30(SMP-30)是同源结构。相应抗体主要只见于HCC病人,在20例其它5种恶性肿瘤和4例肝硬化病人未查到相应抗体,20例慢性肝炎病人血清和20例正常人血清均只见1例呈阳性反应。结论:衰老标记蛋白-30为一种新发现的HCC相关抗原,对HCC有诊断学意义。
Objective:To investigate specific antigens of hepatocelluar carcinoma (HCC) Methods:Serological identification of antigens by recombinant expression cloning (SEREX) technique was used for screening of specific antigens of hepatocelluar carcinoma cDNA library of hepatocellular carcinoma tissues from Guangxi.These antigens reacted to the sera of other persons,and analysis of these clonic gene sequences was conducted Results:One of these antigens--HOM HCC 22 5,was homologous with human mRNA for senescence marker protein 30 (SMP 30) Serologic test demonstrated that 3/5 HCC had positive reaction to HOM HCC 22 5 Twenty sera from different kinds of neoplasm and four from hepatocirrhosis were all negative Conclusion:The results of sera analysis suggest that HOM HCC 22 5/SMP 30 is a new HCC associated antigen and will be of great value to diagnosis of HCC
