摘要
目的建立大鼠尿白蛋白放免测定方法及其应用意义。方法用大鼠白蛋白免疫青紫蓝兔,制备抗血清,采用双抗加PEG法。结果本方法测定范围25~1600μg/L,灵敏度为9.78μg/L,回收率89.2%~112.0%,批内、批间平均变异系数分别为4.1%和8.3%。大鼠在糖尿病成模2周时,24小时尿白蛋白排泄率(UAE)升高,而培哚普利治疗组尿白蛋白与正常大鼠组无明显差异,在治疗6个月后尿白蛋白明显低于糖尿病(P<0.01),培哚普利能明显抑制糖尿病大鼠尿白蛋白排泄,对糖尿病大鼠肾脏有某种保护作用。结论大鼠尿白蛋白放免测定方法的建立为糖尿病大鼠肾病模型和其他肾病的研究工作提供了一个客观的。
Objective To establish a radioimmunoassay method for determination of urinary albumin excretion (UAE) in rats and to evaluate its significance. Methods The rabbits were immunized with rat albumin. Rat urinary albumin excretion was determined radioimmunologically with the prepared antiserum and second antibody PEG technique. Results The measurement range of this second antibody PEG method was 25~1600μg/L. Recoveryrateandintra andinter batchvariation coefficients were 89.2%~112.0%, 4.1% and 8.3% respectively. 24h UAE was measured in rats which were divided into three groups: the diabetic controls (DM group), the group treated with Perindopril (DP group) and the normal controls (NC group). The 24h UAE in DC group was elevated at 2 weeks and increased significantly thereafter, whereas there was no significant difference in 24 h UAE between DP and NC groups, suggesting that Perindopril might have some renal protective effects. Conclusion The establishment of a radioimmunoassay method for UAE might provide a reliable and sensitive means of early diagnosis in experimental diabetic nephropathy.
