摘要
目的观察甘草酸防治糖尿病肾病的作用及其可能机制。方法用链脲佐菌素制备糖尿病大鼠模型(糖尿病模型组);糖尿病甘草酸干预组(甘草酸组)给予甘草酸(100 mg.kg.d-1)灌胃,每天1次。对比观察肾脏病变和肾小球TGF-β1表达。结果肾质量/体质量比值,甘草酸组明显小于糖尿病组(P<0.05)。免疫组化可见,60天时肾小球TGF-β1阳性面积比例,糖尿病组明显高于正常组(P<0.05);而甘草酸组其比例,显著低于糖尿病模型组(P<0.05)。W estern b lot可见,60天时肾脏TGF-β1水平,甘草酸组明显低于糖尿病组(P<0.05)。结论甘草酸对糖尿病时的肾小球具有一定的保护作用,可能与TGF-β1减少有关。
Objective To investigate the effect of glycyrrhizic acid on the diabetic nephroperthy and its mechanism in rats. Methods Glycyr- rhetinic acid, 100 mg · kg-1, was fill with once a day in the diabetes rats model induced by Streptozotocin. The kidney lesions and the expres- sion of TGF - β1 in the glomerulus were comparably observed on the 14, 30 and 60 days after the treatment. Results Compared with the diabetic rats, the ratio of kidney weight / body weight in the glycyrrhizic acid treatment group was significantly decreased ( P 〈 0. 05 ). At the time 60 days, a significant increased expression of TGF- β1 in the glomerulus was observed in the diabetic group by immunohistochemistry. Comparing with the diabetes group, the expression of TGF - β1 in the glycyrrhizin acid group was markedly decreased ( P 〈 0. 05 ). According to Western blot, expression of TGF - β1 of kidney in the glycyrrhizin acid group at 60 days was markedly decreased comparing with diabetic group (P 〈 0. 05 ). Conclusion Glycyrrhizin acid can inhibit the expression of TGF - β1 and protect the glomerulus of this diabetes model in a certain extent.
