高黎贡山温泉嗜热菌的分离及生理特征研究 被引量:3

摘要 [目的]研究高黎贡山温泉一株嗜热菌的生态特征及营养需求特征,为高温微生物菌种资源研究提供参考。[方法]采集高黎贡山百花岭高山温泉水样,通过富集培养和平板划线分离纯化,得到一批嗜热菌。[结果]从分离得到的嗜热菌中选择其中菌落较大、生长势良好的GLGS3菌株进行研究。经测定,GLGS3菌株培养温度为45~65℃时,菌体生长情况较好,最适温度是50℃;GLGS3菌株的培养pH值为7.5~8.5时,菌体生长情况较好,最适pH值是8.0。该菌株的吲哚试验阴性,能分解葡萄糖产酸、产气。同时还能利用果糖、乳糖、麦芽糖、淀粉等多种糖作为碳源生长,能利用有机氮、硝态氮等氮源。[结论]对高黎贡山百花岭地区温泉样品中嗜热微生物生长习性的研究,有助于了解该地区温泉特定生境下可培养微生物群落类型。 [Objective]The reference for the development of the resource of microbial species with the tolerance to high temperature was provided through research on the ecological characteristics of one Thermophilies strain isolated in the Gaoligongshan Mountain Spring and its nutrient need.[Method]A set of Thermophilies strains was collected in the water sample from the Baihualing Spring in Gaolingongshan Mountain after they were isolated by means of the method of enrichment culture and plate streaking.[Results]The GLGS3 that was big colony with rapid growth was chosen for its future research.It was found that GLGS3 grew well in the range of temperature from 45 ℃ to 65 ℃ and the best temperature for its growth was 50 ℃;and the pH value ranges from 7.7 to 8.5 and the best pH value for its growth is 8.0.The indole test of the strain was negative and had the ability in the glucose-decomposing for acid and gas production.Meanwhile,it could use the fructose,lactose,maltose,starch and other sugars as a carbon source and the organic nitrogen,nitrate and others as nitrogen sources for its growth.[Conclusion]This work can help us learn the culturable microbiology’s style of this area.
