千层金组培苗瓶外生根技术研究 被引量:12

摘要 [目的]对千层金瓶外生根技术进行研究。[方法]以千层金健壮试管苗为材料,使用不同浓度生根促进剂NAA、IBA、ABT1、ABT2处理穗条,再测定各处理的扦插生根率。[结果]用浓度100mg/L的ABT1浸泡穗条60min后扦插,生根率为79.3%,再用该药液把滑石粉稀释至糊状,随蘸随插,生根率上升到93.0%,其中,以复合基质A(河沙:珍珠岩:蛭石:泥炭土=1:2:2:5)处理效果最佳,发根时间早,8d后长出新根,平均不定根数量最多,根长最长,分别为4.5条和3.6cm/株。[结论]该研究结果可为建立和完善千层金离体再生技术工厂化生产提供科学依据。 [Objective]The research aimed to study the rooting techniques of Melaleuca bracteata outside test tubes.[Method]Taken healthy test-tube plantlet of M.bracteata as materials,and the plant cuttings were treated with different concentrations of root accelerants such as NAA,IBA,ABT1 and ABT2.And then the rooting rates of different treatments were determined.[Result]The results showed that the average rooting rate was 79.3% when cuttings were socked in aqueous solution of 100 mg/L ABT1 for 60 minutes.Then the liquid medicine was used to dilute talc into paste,and the cuttings were planted after dipped in the paste.Thus,the rooting rate rose to 93.0%.Among all treatments,the one with complex substrate such as sphagnum:perlite:vermiculite:peat soil=1:2:2:5 had optimum effects,and the rooting time is earlier,new roots appeared after 8 days.The average number of adventitious root and root length were 4.5 pieces and 3.6 cm.[Conclusion]The results of this research provided scientific basis for the establishment and improvement of the industrial production of M.bracteata regeneration technology.
