摘要 目的探讨阿仑磷酸钠对大鼠成骨细胞代谢调控的影响。方法取新生SD大鼠的颅骨,胶原酶法培养成骨细胞。用MTT、ALP检测和Western Blot方法观察阿仑磷酸钠对体外培养成骨细胞的增殖、碱性磷酸酶活性和OPG、RANKL蛋白表达的影响。结果阿仑磷酸钠对成骨细胞增殖无明显影响,但能提高成骨细胞碱性磷酸酶活性,增强OPG蛋白的表达,降低RANKL蛋白的表达(P<0.05或P<0.01)。结论阿仑磷酸钠能促进成骨细胞的分化,并通过调节OPG/RANKL蛋白比值影响骨代谢。 【Objective】To explore the effect of Alen dronate on the osteoblasts metabolism in rats in vitro.【Methods】Osteoblasts were obtained from new born rat calvaria by digestive enzymes.MTT,ALP Detection and Western Blot were used to observe the proliferation,activity of ALP and protein expression of OPG and RANKL of cultured osteoblasts in vitro.【Results】Alendronate had no effect on stimulating cell proliferation,alendronate promoted the activity of ALP,alendronate promoted the protein expression of OPG and inhibited the protein expression of RANKL of cultured osteoblasts(P 0.01 or P 0.05).【Conclusion】Alendronate can promote differentiation,and may influence the bone metabolism by regulating the OPG/RANKL protein expression.
